Neurotrophic Factor Secretion

Neurotrophic factor secretion is the regulated release of proteins that support neuronal survival, growth, differentiation, and synaptic function. Neurons and glial cells produce factors such as nerve growth factor and brain-derived neurotrophic factor, which are released in response to cellular activity or injury and activate receptors on nearby or connected cells through paracrine or retrograde signaling. This process shapes neural development, maintains circuit function, and contributes to repair after nervous-system damage. Studying neurotrophic factor secretion helps clarify mechanisms of synaptic plasticity and neurodegeneration while informing research on neuroprotective strategies, regenerative therapies, and neurological disease models.

This article illustrates the use of pulse-chase radio labeling in combination with site-specific photocrosslinking to monitor interactions between a protein of interest and other factors in E. coli. Unlike traditional chemical cross-linking methods, this approach generates high resolution “snapshots” of an ordered assembly pathway in a living cell.

We describe a matrigel plug assay to illustrate angiogenic potential of a pool of factors secreted by cancer cells using two complementary imaging modalities, ultrasound and endomicroscopy. The matrigel, an extracellular matrix (ECM)-mimic gel, is utilized to introduce the host (mouse) with angiogenic factors secreted to the conditioned media...

Neuron-Macrophage Co-cultures to Activate Macrophages Secreting Molecular Factors with Neurite Outgrowth Activity

The current protocol presents experimental procedures to stimulate cultured macrophages to be endowed with capacity to release molecular factors that promote neurite outgrowth. Treatment of cAMP to the neuron-macrophage co-cultures induces the macrophages to produce conditioned medium that possesses strong neurite outgrowth activity.

Here, we propose three different methods of damaging the sensory fibres innervating the cornea. These methods facilitate the study of axon regeneration in mice. These three methods, which are adaptable to other animal models, are ideal for the study of corneal innervation physiology and regeneration.
