Subependymal Npc Isolation

Subependymal NPC isolation is a method for extracting neural progenitor cells from the subependymal zone lining the lateral ventricles, a region associated with adult neurogenesis. The procedure typically involves dissecting the subependymal tissue, enzymatically and mechanically dissociating it into a single-cell suspension, and culturing the cells with mitogens such as epidermal growth factor and fibroblast growth factor to promote progenitor expansion and neurosphere formation. Isolated cells provide an experimental model for studying neural stem cell maintenance, proliferation, differentiation, and migration, while supporting research on brain development, injury repair, neurodegenerative disease, and regenerative strategies.

Cryo-section-dissection allows fresh, frozen preparation of the largest neurogenic niche in the murine brain for deep quantitative proteome analysis. The method is precise, efficient, and causes minimal tissue perturbation. Therefore, it is ideally suited for studying the molecular microenvironment of this niche, as well as other organs, regions, and species.

Isolation and Culture of Mouse Kupffer Cells

In this video, we demonstrate the isolation of Kupffer cells or liver macrophages from the mouse liver. The liver is perfused with a digestion solution followed by density gradient separation. The isolated adherent Kupffer cells can be used for further analysis.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.
