Biotinylated Metabolite

A biotinylated metabolite is a metabolic compound or intermediate that contains biotin covalently attached to its structure, enabling specialized biochemical reactions. In biotin-dependent carboxylation, ATP activates bicarbonate, and the resulting carboxyl group is transferred to the ureido ring of biotin before biotin carries it between catalytic sites or onto a substrate. These compounds are central to pathways involving fatty acid synthesis, amino acid metabolism, and energy production. Studying biotinylated metabolites helps clarify enzyme mechanisms, metabolic regulation, and the biochemical consequences of impaired biotin utilization, with relevance to nutrition, inherited metabolic disorders, and metabolic research.

Here, we detail the method of Sequencing of Psoralen crosslinked, Ligated, and Selected Hybrids (SPLASH), which enables genome-wide mapping of intramolecular and intermolecular RNA-RNA interactions in vivo. SPLASH can be applied to study RNA interactomes of organisms including yeast, bacteria and humans.

The protocol presents a method for in vivo covalent attachment of biotin to proteins based on their proximity to a biotin ligase fused to a protein of interest. This modification allows for a selective enrichment of the proteins using streptavidin beads as needed in protein interaction studies.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.
