Cholesterol Modified Oligonucleotides

Cholesterol modified oligonucleotides are short DNA or RNA strands covalently linked to cholesterol to alter their physicochemical behavior and improve performance in biological systems. The hydrophobic cholesterol group gives the conjugate amphiphilic character, promoting association with lipid membranes and influencing cellular uptake, transport, and distribution, while the oligonucleotide retains sequence-specific binding to nucleic acid targets. In bioengineering, these conjugates support the delivery and study of antisense oligonucleotides, small interfering RNAs, and other gene-regulating molecules. Their membrane interactions can improve localization and stability, helping researchers design more effective tools for gene silencing, molecular diagnostics, and therapeutic development.

Here, a protocol for assaying nucleic acid metabolizing enzymes is presented, using examples of ligase, nuclease, and polymerase enzymes. The assay utilizes fluorescently labeled and unlabeled oligonucleotides that can be combined to form duplexes mimicking RNA and/or DNA damages or pathway intermediates, allowing for the characterization of enzyme behavior.

Macrophage Cholesterol Depletion and Its Effect on the Phagocytosis of Cryptococcus neoformans

In this article, a protocol for infection of macrophages with Cryptococcus neoformans is described. Also, a method for sterol depletion from the macrophages is explained. These protocols provide a guide to study fungal infections in vitro and examine the role of sterols in such infections.
