Drug Loading Efficiency

Drug loading efficiency is a measure of how much therapeutic compound becomes incorporated into a drug delivery system relative to the amount used during its preparation. It is typically calculated as the mass of drug successfully encapsulated or adsorbed by a carrier, divided by the initial drug mass, with performance influenced by drug–material interactions, carrier composition, and processing conditions. In bioengineering, this metric helps researchers compare nanoparticles, microparticles, hydrogels, and other delivery platforms, optimize formulation procedures, and predict dose consistency, release behavior, and therapeutic performance. Improving loading efficiency can also reduce drug waste and support more effective, scalable delivery systems.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.
