Fluo-4 Dose

Fluo-4 dose refers to the amount or working concentration of the fluorescent calcium indicator used to measure intracellular calcium dynamics, an important parameter in cellular bioengineering and live-cell imaging. After cell loading, the membrane-permeant Fluo-4 AM ester enters cells and is cleaved by intracellular esterases, releasing Fluo-4, whose fluorescence increases when it binds calcium ions. Selecting an appropriate dose balances signal intensity, background fluorescence, loading efficiency, and potential effects on cell function. Optimized dosing supports reliable calcium imaging in engineered tissues, biosensors, and cell-based assays, enabling analysis of signaling, contraction, and responses to mechanical or chemical stimulation.
