3d Human Skin Reconstruct

A 3D human skin reconstruct is a laboratory-engineered tissue model that reproduces key structural and functional features of human skin, offering a controlled alternative to studying intact tissue. It typically combines human keratinocytes and fibroblasts with a collagen-based or other biomaterial scaffold, then uses culture conditions such as an air-liquid interface to promote dermal organization and epidermal stratification. In bioengineering, these models support research on skin development, barrier function, wound healing, disease mechanisms, and responses to pharmaceuticals or cosmetic ingredients. Their controlled composition and reproducibility can reduce reliance on animal testing while advancing engineered skin and regenerative medicine.

In this report, we describe the three-dimensional skin reconstruct model which mimics human skin in architecture and composition. Melanocyte physiology, melanoma progression and the fate of dermal stem cells have been investigated using the skin reconstruct model. The model is also useful as a preclinical tool for drug assessment.
