Polyglutamine Aggregation

Polyglutamine aggregation is the misfolding and assembly of proteins containing abnormally expanded glutamine-rich sequences, a process central to several inherited neurodegenerative disorders. Expansion of CAG trinucleotide repeats produces elongated polyglutamine tracts that destabilize protein structure, promote intermolecular interactions, and drive the formation of soluble oligomers and insoluble amyloid-like fibrils. Biological techniques such as protein-expression systems, fluorescence assays, microscopy, and biochemical aggregation measurements help characterize these species and their effects on cells. Studying polyglutamine aggregation clarifies mechanisms of neuronal toxicity and supports evaluation of genetic, pharmacological, and molecular strategies designed to reduce misfolding or enhance aggregate clearance.

This manuscript describes three complementary protocols for assessing the toxicity of polyglutamine (polyQ)-expansion proteins in the yeast Saccharomyces cerevisiae. These protocols can easily be modified to monitor the toxicity of other misfolded proteins in yeast.
