Cardiomyocyte Transduction

Cardiomyocyte transduction is the process of introducing genetic material into heart muscle cells to alter or study gene expression, making it an important technique in cardiovascular biology. Typically, viral or nonviral vectors deliver DNA, RNA, or genome-editing components across the cardiomyocyte membrane, after which intracellular trafficking enables the cargo to reach its site of action and produce a biological effect. Researchers use transduction to investigate cardiac gene function, model inherited and acquired heart disease, test therapeutic strategies, and support studies of cardiac regeneration. Efficient, selective delivery can improve experimental reliability and advance the development of gene-based treatments.

The following protocol describes the isolation and culture of primary cardiomyocytes from neonatal mouse hearts. The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.
