Ba/f3 Cell Line

The Ba/F3 cell line is a murine pro-B cell model that requires interleukin-3 (IL-3) for survival and proliferation, making it useful for studying cytokine-dependent growth and oncogenic signaling. In culture, IL-3 activates receptor-associated signaling pathways that support cell expansion; introducing an activated kinase or oncogenic fusion can drive proliferation without IL-3, creating a measurable transformation phenotype. Researchers use Ba/F3 cells to assess kinase function, evaluate mutation-dependent drug sensitivity, and characterize targeted inhibitors by comparing growth in IL-3-dependent and cytokine-independent conditions. This system provides a practical link between molecular signaling mechanisms and therapeutic response.

Assessing Cell Viability and Death in 3D Spheroid Cultures of Cancer Cells

Here, we present several simple methods for evaluating viability and death in 3D cancer cell spheroids, which mimic the physico-chemical gradients of in vivo tumors much better than the 2D culture. The spheroid model, therefore, allows evaluation of the cancer drug efficacy with improved translation to in vivo conditions.

A Simple Bioassay for the Evaluation of Vascular Endothelial Growth Factors

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
