Culture-dependent Methods

Culture-dependent methods are biological techniques that detect, isolate, and characterize microorganisms by growing them under controlled laboratory conditions, making them valuable for studying viable populations. In practice, a sample is inoculated onto or into a suitable growth medium and incubated with defined nutrients, temperature, oxygen levels, and time; resulting colonies or enrichment cultures can then be counted, purified, and identified using phenotypic or molecular tests. These methods support microbial isolation, antimicrobial susceptibility testing, food and water quality assessment, and clinical diagnosis. However, they may underestimate diversity because some organisms require specific conditions or do not grow in standard culture media.

A High Resolution Method to Monitor Phosphorylation-dependent Activation of IRF3

Here we describe a procedure allowing a detailed analysis of the phosphorylation-dependent activation of the IRF3 transcription factor. This is achieved through the combination of a high resolution SDS-PAGE and a native-PAGE coupled to immunoblots using multiple phosphospecific antibodies.

Methods for the manipulation and analysis of NF-κB-dependent adult hippocampal neurogenesis are described. A detailed protocol is presented for a dentate gyrus-dependent behavioral test (termed the spatial pattern separation-Barnes maze) for the investigation of cognitive outcome in mice. This technique should also help enable investigations in other experimental settings.

Method for Culture of Early Chick Embryos ex vivo (New Culture)

This video demonstrates New culture, a method by which chick embryos are cultured outside the egg for up to 24 hr. This method enables one to study early development (primitive streak to 14 som.), a period corresponding to E7-9 in mouse. Applications of this technique include electroporation, in situ hybridization and immunohistochemistry.

In this video protocol, we demonstrate our standard procedures of rat whole embryo culture after E12.5 using the rotator-type bottle culture system.
