Drosophila Preparation

Drosophila preparation is the controlled collection, handling, and setup of fruit flies for laboratory experiments, enabling researchers to examine development, genetics, physiology, and behavior. During preparation, flies are typically immobilized with carbon dioxide or brief cooling, then sorted by strain, sex, age, or visible phenotype and transferred to experimental containers or microscope slides. Careful handling limits stress, injury, and unwanted variation that could affect results. Prepared Drosophila support genetic crosses, behavioral assays, anatomical imaging, and studies of conserved biological processes, making this model organism useful for investigating gene function and disease mechanisms.

Views of live cultures show cells 1 hour after plating and differentiated neurons after 2 days of growth in a bicarbonate-based defined medium. The neurons are electrically excitable and form synaptic connections.

Preparation of Adult Drosophila Eyes for Thin Sectioning and Microscopic Analysis

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.
