Germinating Seeds

Germinating seeds are seeds undergoing the biological transition from dormancy to active growth, a process essential for plant reproduction and establishment. When a seed absorbs water through imbibition, its tissues swell, metabolism resumes, stored nutrients are mobilized, and cell division and elongation begin; under suitable conditions of moisture, oxygen, and temperature, the embryonic radicle emerges first. Studying seed germination helps explain plant development, dormancy, and responses to environmental conditions. It also supports agriculture by improving crop establishment, seed testing, propagation methods, and selection of varieties suited to changing climates.

We present the procedure to assemble a seed coat bedding assay (SCBA) from Arabidopsis thaliana seeds. The SCBA was shown to be a powerful tool to explore genetically and in vitro how the endosperm controls seed germination in dormant seeds and in response to light cues. The SCBA is in principle applicable under any situation where the endosperm is suspected to influence embryonic growth.

Here, we present a protocol to optimize asymbiotic seed germination, seedling development, and shoot elongation in Hemipilia cucullata. The protocol also enables the induction of protocorm-like bodies from sterile leaf explants, facilitating efficient in vitro propagation for the conservation and sustainable use of this endangered medicinal orchid.
