Gut Protozoa Extraction

Gut protozoa extraction is the laboratory process of recovering single-celled eukaryotic organisms from gastrointestinal contents or fecal material for biological analysis. It typically uses sample collection followed by suspension, filtration, sedimentation, or other separation steps that concentrate protozoa while removing larger debris and unwanted material; preservation or culture may then support further study. Extracted organisms can be examined by microscopy, identified through morphological features or molecular methods, and assessed for abundance, diversity, and interactions with their hosts. In biology, this approach supports research on gut microbial communities, host health, disease, ecology, and the roles of protozoa in gastrointestinal ecosystems.

We present procedures for demonstrating that ligands bind to the surface membrane of the cellulose-digesting protozoa in the gut of Formosan subterranean termites using fluorescent microscopy and that ligands coupled with lytic peptides kill these protozoa in vitro (anaerobic protozoa culture) and in vivo (injection into the termite hindgut).

This video illustrates the technique for extracting DNA from the species of microbes resident in the termite hindgut. The preparation of a wet mount slide, which is useful for visualizing the gut microbial community is also illustrated, and a tour through the species-rich gut environment is given.

Oral Infection Alters the Gut Microbiota in a Rat Model

Begin with a rat model with sutures tied around its upper molar teeth to support bacterial colonization.Repeatedly smear the sutures with a gum disease-causing oral pathogenic bacterium using a gel-like solution. The bacteria colonize the gum around the sutures.Colonization triggers inflammation and damages the tissues. The bacteria release toxins that diffuse through damaged tissues and enter the bloodstream. These circulating toxins reach the gut, cause inflammation, disrupt the epithelial...
