Heterozygous Offspring

Heterozygous offspring are individuals that inherit two different alleles for a particular gene, a genetic condition that contributes to variation within biological populations. They arise when each parent contributes a different allele, producing a genotype in which one allele may be dominant while the other is recessive, or both may influence the observable trait through incomplete dominance or codominance. Identifying heterozygous offspring helps researchers and students connect genotype with phenotype, predict inheritance patterns using genetic crosses, and track how traits are transmitted across generations. This concept is central to classical genetics, breeding studies, and the analysis of inherited characteristics.

Novel strategies to faithfully model somatic mutations in hematopoietic stem and progenitor cells (HSPCs) are necessary to better study hematopoietic stem cell biology and hematological malignancies. Here, a protocol to model heterozygous gain-of-function mutations in HSPCs by combining the use of CRISPR/Cas9 and dual rAAV donor transduction is described.

Maternal infection is a risk factor for neurodevelopmental disorders. Mouse models of maternal immune activation (MIA) may elucidate infection's impact on brain development and function. Here, general guidelines and a procedure are provided to produce reliably resilient and susceptible offspring exposed to MIA.

Measuring Mitochondrial Electron Transfer Complexes in Previously Frozen Cardiac Tissue from the Offspring of Sow: A Model to Assess Exercise-Induced Mitochondrial Bioenergetics Changes

Preparation of mitochondria-enriched samples from previously frozen archived solid tissues allowed the investigators to perform both functional and analytical assessments of mitochondria in various experimental modalities. This study demonstrates how to prepare mitochondria-enriched preparations from frozen heart tissue and perform analytical assessments of mitochondria.
