Insulin Secretion Analysis

Insulin secretion analysis is the measurement and interpretation of insulin release from pancreatic beta cells, tissues, or organisms to assess glucose regulation and beta-cell function. In a typical glucose-stimulated assay, elevated glucose increases cellular ATP, closes ATP-sensitive potassium channels, depolarizes the beta-cell membrane, and opens voltage-gated calcium channels; the resulting calcium influx triggers insulin-granule exocytosis. Researchers use techniques such as immunoassays and dynamic secretion measurements to compare insulin responses under different conditions. These analyses support studies of diabetes, metabolic physiology, beta-cell biology, and therapeutic compounds that alter insulin release.

Transcellular protein interactions are important determinants of pancreatic beta-cell function. Detailed here is a method—adapted from a coculture model of synaptogenesis—for investigating how specific transmembrane proteins influence insulin secretion. Transfected HEK293 cells express proteins of interest; beta cells do not need to be transfected or otherwise directly perturbed.

This protocol describes how to perform rapid low-cost luciferase assays at medium-throughput using an insulin-linked Gaussia luciferase as a proxy for insulin secretion from beta cells. The assay can be performed with most luminescence plate readers and multichannel pipettes.

Epigenetic Conversion as a Safe and Simple Method to Obtain Insulin-secreting Cells from Adult Skin Fibroblasts

Here, a new method that allows the conversion of adult skin fibroblasts into insulin-secreting cells is presented. This technique is based on epigenetic conversion, does not involve the use of retroviral vectors nor the acquisition of a stable pluripotent state. It is therefore highly promising for translational medicine applications.
