Organoid Proliferation

Organoid proliferation is the expansion of three-dimensional, organ-like cell cultures through coordinated cell growth and division, providing a tractable model of tissue development and maintenance. In a controlled culture environment, stem or progenitor cells self-renew and produce differentiated descendants while cell-cell interactions and tissue architecture support continued organoid growth. Measuring proliferation helps researchers assess organoid viability, maturation, and responses to changes in culture conditions or experimental treatments. In biology, these models support studies of development, disease mechanisms, drug effects, and regenerative processes, while controlled expansion enables the generation of consistent material for imaging, molecular analysis, and comparative experiments.

The video demonstrates a method for generating human brain organoids from induced pluripotent stem cells. It shows how the neurosphere forms a neuroectodermal layer, which suppresses the formation of other germ layers. The neurospheres are embedded in a 3D gel matrix and form neuroepithelial buds. When incubated with organoid media, these neuroepithelial buds differentiate into specific brain regions, resulting in the formation of mature brain organoids.
