Phage Display Technology

Phage display technology is a molecular screening method that links a peptide or protein’s function to the DNA sequence encoding it, enabling rapid identification of binding molecules. In this approach, bacteriophages are genetically engineered to present peptides, antibody fragments, or proteins on their surface while carrying the corresponding genes inside; libraries are exposed to an immobilized target, and bound phages are recovered, amplified in bacteria, and enriched through repeated selection cycles. The method supports antibody discovery, protein engineering, epitope mapping, and studies of molecular recognition. By connecting genotype with phenotype, phage display accelerates the development of research reagents, diagnostics, and potential therapeutics.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

Phage display is a powerful technique to capture proteins or protein moieties that interact with an immobilized molecule of interest. Once a decision of the type of phage cDNA library to create and screen has been made, the protocol described here permits efficient affinity selection leading to identification of interactors.
