Protein Dimer Elution

Protein dimer elution is the recovery of noncovalent or covalent protein dimers from a separation medium, an important step for isolating and characterizing defined oligomeric states. In chromatography, dimers can be separated from monomers or larger complexes according to properties such as hydrodynamic size, affinity for a ligand, or charge; elution conditions then release the bound or resolved species while preserving, or intentionally disrupting, dimer formation. Researchers use protein dimer elution to assess assembly, stability, purity, and biological activity, supporting studies of enzyme function, receptor signaling, structural biology, and protein engineering.

This manuscript describes an approach to isolate select cognate RNPs formed in eukaryotic cells via a specific oligonucleotide-directed enrichment. We demonstrate the applicability of this approach by isolating a cognate RNP bound to the retroviral 5' untranslated region that is composed of DHX9/RNA helicase A.

High-Performance Liquid Chromatography: Elution Process

2024

In High-Performance Liquid Chromatography (HPLC), the elution process is critical to the separation of analytes and the quality of chromatographic results. Elution describes how compounds move through the column and separate based on their interactions with the mobile and stationary phases. This process determines the resolution, peak shape, and retention times in the chromatogram, which are essential for identifying and quantifying components in complex mixtures. Understanding the elution...

Here, we present a protocol to evaluate the antibacterial efficacy of an antibiotic-eluting polymer to simulate prophylactic clinical application by using a commercially available real-time ATP-based luminescent microbial viability assay. This method enables the monitoring of the longitudinal activity of drug-eluting materials and can be widely adapted to test anti-microbial drug delivery platforms.
