Retinal Whlemount Preparation

Retinal wholemount preparation is a tissue-processing technique that preserves the retina as a flat sheet for examining its cellular organization across the entire tissue. The method involves carefully dissecting the retina from the eye, making strategic radial cuts to relieve curvature, and mounting it photoreceptor or ganglion-cell side up for staining and microscopy. This approach enables researchers to map retinal layers, neuronal populations, blood vessels, and regional patterns without relying on limited tissue sections. In biology, retinal wholemounts support studies of development, neurodegeneration, vascular disease, regeneration, and experimental treatments by revealing spatial changes across the complete retinal surface.
