Dye Diffusion Assay

A dye diffusion assay is a laboratory method that measures how a colored solute spreads through a medium, providing a simple model of molecular transport in biological systems. The dye moves by random molecular motion from regions of higher concentration to lower concentration, while diffusion rate depends on factors such as concentration gradient, temperature, distance, and the properties of the surrounding material. Researchers and students use this assay to investigate passive transport, compare diffusion through gels or membranes, estimate permeability, and examine how physical conditions influence molecular movement. Its results help connect observable color changes with fundamental processes in cell biology and physiology.

This protocol describes a simple, cost effective way to individually identify Drosophila or other insects. Demonstration data investigating mating success across three species of Drosophila show that this method is comparable or better than the use of CO2 anaesthesia.

Cell Membranes and Diffusion In order to function, cells are required to move materials in and out of their cytoplasm via their cell membranes. These membranes are semipermeable, meaning that certain molecules are allowed to pass through, but not others. This movement of molecules is mediated by the phospholipid bilayer and its embedded proteins, some of which act as transport channels for molecules that otherwise would not be able to pass through the membrane, such as ions and carbohydrates.
