Hippo Pathway Screening

Hippo pathway screening is the systematic testing of genes, proteins, compounds, or cellular conditions that alter the Hippo signaling network, a growth-control system important in cancer biology. In this pathway, MST1/2 and LATS1/2 kinases phosphorylate YAP and TAZ, promoting their cytoplasmic retention or degradation; when signaling is reduced, YAP/TAZ enter the nucleus, bind TEAD transcription factors, and activate genes that support proliferation and survival. Screening assays measure these changes through reporter activity, protein localization, phosphorylation, or cell growth. The results help identify pathway regulators, clarify mechanisms of tumor development, and support therapeutic discovery targeting YAP/TAZ or related signaling components.

Here, we describe a methodology to perform a targeted siRNA “ubiquitome” screen to identify novel ubiquitin and ubiquitin-like regulators of the HIF1A-mediated cellular response to hypoxia. This can be adapted to any biological pathway where a robust read out of reporter activity is available.

Bacterial pathogens secrete proteins into the host that target crucial biological processes.
