Immunocompromised Mouse

An immunocompromised mouse is a laboratory mouse with weakened or absent immune functions, making it useful for studying human cells and tissues in vivo. Genetic alterations, such as defects in T cells, B cells, or innate immune pathways, and sometimes conditioning treatments reduce immune rejection, allowing implanted human tumors to establish and grow. In cancer research, these mice support xenograft and patient-derived tumor models for investigating tumor growth, metastasis, therapeutic response, and drug efficacy. Because immune deficiencies can alter tumor-host interactions, researchers often complement these models with humanized or immunocompetent mice when evaluating immune-based therapies and translational relevance.

Intrafemoral injections allow for the engraftment of a small number of hematopoietic stem and progenitor cells (HSPCs), by placing the cells directly in the bone marrow cavity. Here we describe an experimental protocol of intrafemoral injection of human HSPCs into immunodeficient mice.
