Immunomagnetic Cell Separation

Immunomagnetic cell separation is a laboratory method that isolates specific cell populations from mixed samples by combining antibody-based recognition with magnetic particle handling. In positive selection, antibodies attached to magnetic beads bind target surface markers, and a magnetic field captures labeled cells while unlabeled cells are removed; negative selection instead removes unwanted populations to enrich the remaining cells. In cancer research, this approach can enrich tumor cells or immune subsets from blood or tissue, supporting flow cytometry, molecular profiling, culture, and functional assays. By improving sample purity and reducing manual separation steps, it helps researchers characterize tumor heterogeneity, investigate immune responses, and obtain defined populations for translational studies.

We present the techniques required to isolate the stromal vascular fraction (SVF) from mouse inguinal (subcutaneous) and perigonadal (visceral) adipose tissue depots to assess their gene expression and collagenolytic activity. This method includes the enrichment of Sca1high adipose-derived stem cells (ASCs) using immunomagnetic cell separation.

2023

The video explains a way to purify B-cells by immunomagnetic targeting and separation from peripheral blood mononuclear cells suspension. The method involve labeling unwanted cell types with specific antibodies or ligands (linked to magnetic particles) targeting specific cell surface proteins, to be depleted from the heterogenous cell suspension by magnetic separation.
