Nsg Mouse Model

The NSG mouse model is a highly immunodeficient laboratory mouse used to study human cells, tissues, and tumors in vivo, making it valuable in cancer research. It combines the NOD background with mutations in Prkdc and Il2rg, which prevent functional T- and B-cell development and severely impair natural killer cell activity, allowing human tumor cells or patient-derived xenografts to engraft. Researchers use NSG mice to investigate tumor growth, metastasis, treatment response, and interactions between cancer cells and the human immune system. These models support preclinical evaluation of targeted therapies, immunotherapies, and personalized cancer treatments while helping assess biological outcomes in a living organism.

This protocol provides a method to establish humanized mice (hu-NSG) via intrahepatic injection of human hematopoietic stem cells into radiation-conditioned neonatal NSG mice. The hu-NSG mouse is susceptible to HIV infection and combinatorial antiretroviral therapy (cART) and serves as a suitable pathophysiological model for HIV replication and latency investigations.

In this video, human-derived melanoma cells are implanted subcutaneously into the flank region of an immunocompromised mouse. The PDX model allows preclinical investigation of melanoma cells that better recapitulate the tumor heterogeneity and melanoma aggressiveness observed in in vivo conditions.

Mouse Models of Periventricular Leukomalacia

We established mouse models of periventricular leukomalacia (PVL), the predominant brain injury in premature infants characterized by periventricular white matter lesions. Hypoxia/ischemia with/without systemic infection are the primary causes of PVL. Unilateral carotid ligation and hypoxia exposure with/without lipopolysaccharide injection creates PVL-like lesions in P6 mice.

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.
