Peroxidase Blocking

Peroxidase blocking is a sample-preparation step that suppresses endogenous peroxidase activity before antibody-based detection, helping distinguish a true cancer-associated signal from nonspecific background. In immunohistochemistry, tissue sections are treated with a blocking reagent, commonly hydrogen peroxide, which inactivates native peroxidase enzymes that could otherwise react with the substrate used to visualize horseradish peroxidase-conjugated antibodies. This step improves contrast and interpretation when identifying tumor markers, immune-cell populations, or signaling proteins in cancer specimens. Effective blocking supports more reliable comparisons among samples, while excessive treatment can reduce assay sensitivity or damage tissue epitopes.
