Endobronchial Biopsy

Endobronchial biopsy is a minimally invasive technique that removes small tissue samples from lesions or abnormal mucosa lining the bronchial airways, providing material for diagnosis and cancer research. During bronchoscopy, a flexible or rigid instrument is guided through the airway, and biopsy forceps collect tissue under direct visualization; the specimen is then preserved for microscopic and, when appropriate, molecular analysis. In lung cancer studies, these samples help identify malignant changes, characterize tumor morphology and biomarkers, evaluate treatment response, and compare tumor tissue with surrounding airway samples. The method links airway-based sampling with clinically relevant evidence for tumor classification and precision-oriented investigation.

Substernal thyroid lesions are common, and need to be differentiated from malignancy. Obtaining percutaneous fine needle biopsy is not possible due to its retrosternal location. This article proposes a protocol for biopsy of substernal thyroid lesions using Endobronchial Ultrasound-guided Transbronchial Needle Aspiration (EBUS-TBNA).

Human Lung Dendritic Cells: Spatial Distribution and Phenotypic Identification in Endobronchial Biopsies Using Immunohistochemistry and Flow Cytometry

Lung-resident immune cells, including dendritic cells (DCs) in humans, are critical for defense against inhaled pathogens and allergens. However, due to the scarcity of human lung tissue, studies are limited. This work presents protocols to process human mucosal endobronchial biopsies for studying lung DCs using immunohistochemistry and flow cytometry.

Systematic Endobronchial Ultrasound - The Six Landmarks Approach

Endobronchial ultrasound-guided sampling using transbronchial needle aspiration plays a key role in staging and diagnosing lung cancer. We propose a systematic stepwise approach dividing the procedure into six landmarks that should be taught to new operators.

Using the Endoscope for Endobronchial Ultrasound in the Esophagus

Transesophageal ultrasound (EUS-B) is a safe and feasible procedure using the echoendobronchoscope (EBUS) in esophagus and stomach. After identifying six anatomical landmarks, additional structures can be identified and biopsied, sparing subsequent diagnostic sessions. Thus, EUS-B is an ideal continuation of bronchoscopy and EBUS in diagnosing lung cancer and other diseases.

The liquid biopsy has revolutionized our approach to oncology translational studies, with sample collection, quality, and storage being crucial steps for its successful clinical application. Here we describe a standardized and validated protocol for downstream circulating-free DNA applications that can be applied in most translational research laboratories.
