Renal Cortex Preparation

Renal cortex preparation is the process of isolating and processing the kidney’s outer cortical tissue for controlled laboratory analysis, a critical step in studying renal structure, function, and disease. It typically involves separating the cortex from the capsule and medulla, then trimming, sectioning, homogenizing, or dissociating the tissue to preserve relevant cellular and molecular features for downstream assays. In cancer research, prepared renal cortex samples support histology, immunostaining, molecular profiling, and cell-based studies of kidney tumors and their surrounding microenvironment. Consistent preparation improves sample quality, enables comparisons between normal and tumor-associated tissue, and strengthens investigations of disease mechanisms and therapeutic responses.

This protocol describes decellularization of Sprague Dawley rat kidneys by antegrade perfusion of detergents through the vasculature, producing acellular renal extracellular matrices that serve as templates for repopulation with human renal epithelial cells. Recellularization and use of the resazurin perfusion assay to monitor growth is performed within specially-designed perfusion bioreactors.
