Tissue Deparaffinization

Tissue deparaffinization is the laboratory process of removing paraffin wax from formalin-fixed, paraffin-embedded tissue sections so their cellular structures and biomolecules become accessible for analysis. Typically, an organic clearing solvent dissolves the wax, followed by graded alcohols and water that rehydrate the specimen and prepare it for staining or molecular treatment. In cancer research, effective deparaffinization supports hematoxylin and eosin histology, immunohistochemistry, and selected nucleic-acid assays used to characterize tumors, detect biomarkers, and compare tissue samples. Careful control of solvent exposure and rehydration helps preserve morphology and assay quality.
