Relapsing Fever

Relapsing fever is an infectious disease caused by Borrelia bacteria and characterized by recurring episodes of fever separated by symptom-free intervals. The pattern results from antigenic variation, in which the bacteria repeatedly alter their surface proteins, evade existing antibodies, and multiply again in the bloodstream; transmission occurs primarily through soft ticks or body lice, depending on the Borrelia species. In clinical practice, recognizing recurrent fever with headache, chills, myalgia, and possible exposure to vectors supports diagnosis through blood-smear examination, molecular testing, or serology. Prompt antimicrobial treatment is important because untreated infection can cause systemic complications and may trigger a Jarisch–Herxheimer reaction.

In vitro culture is a direct detection method for the presence of living bacteria. This protocol describes methods for the culture of diverse Borrelia spirochetes, including those of the Borrelia burgdorferi sensu lato complex, relapsing fever Borrelia species, and Borrelia miyamotoi. These species are fastidious and slow growing but can be cultured.
