Beta Cell Differentiation

Beta cell differentiation is the developmental process by which pancreatic progenitor cells become mature insulin-producing beta cells, a key event in establishing glucose regulation. During development, coordinated signaling and transcriptional programs activate factors such as PDX1, NGN3, NEUROD1, and MAFA, guiding endocrine commitment, beta cell specification, and functional maturation. In developmental biology, studying this process clarifies how cellular identity and pancreatic tissue organization arise. It also supports efforts to generate beta cells from pluripotent stem cells for diabetes research, evaluate mechanisms of beta cell failure, and develop regenerative or cell-replacement therapies.

The present protocol describes an enhanced method to increase the co-expression of PDX1 and NKX6.1 transcription factors in pancreatic progenitors derived from human pluripotent stem cells (hPSCs) in planar monolayers. This is achieved by replenishing the fresh matrix, manipulating cell density, and dissociating the endodermal cells.

Cell-mediated lymphocytotoxicity (CML) assays can be used to test autoreactive responses and study mechanisms of cell death in vitro. However, using live-cell confocal microscopic imaging techniques with fluorescent dyes, the type and kinetics of cell death as well as the pathways utilized can be studied in greater detail.

Beta

2024

Beta is a crucial metric in finance used to assess an investment's risk and expected return relative to the overall market. It is calculated as the slope of the line in a scatterplot that compares the returns of an individual stock to the returns of the market benchmark, typically represented by a broad market index. Beta quantifies how much a stock's price moves in response to market movements, providing insight into its volatility and systematic risk. A beta of one indicates that the stock's...

Imaging Transgenic Beta-Cells in an Immunodeficient Mouse Challenged with Diabetic Splenocytes

This video demonstrates an imaging-based method to visualize transplanted luciferase-expressing beta cells into a non-obese diabetic immunodeficient mouse. The introduction of splenocytes from a diabetic mouse leads to progressive beta cell destruction, visually confirmed by declining luminescence after substrate introduction.
