Neuron Integration

Neuron integration is the process by which a neuron combines incoming excitatory and inhibitory signals to determine whether it will generate an electrical response, making it central to communication within developing neural circuits. At the cell body and axon initial segment, synaptic inputs undergo spatial and temporal summation: their combined effects shift the membrane potential toward or away from the threshold for action-potential initiation. In developmental biology, studying neuron integration reveals how growing neurons refine synaptic connections, balance excitation and inhibition, and establish functional circuits. These principles help explain nervous-system maturation and provide a framework for investigating developmental disorders that disrupt neural connectivity.

2026

This protocol provides an integrated method that includes viral delivery, optrode implantation, photothrombotic induction, simultaneous recording of astrocyte and neuronal activity, and optogenetic stimulation of astrocytes. This integrated method aims to provide concurrent investigation of the astrocyte-neuron relationship across the acute-to-chronic stages of ischemic stroke.

We demonstrate the use of the gene gun to introduce fluorescent dyes, such as DiI, into neurons in brain slices from rodents and non-human primates of different ages. In this particular case, we use adult mice (3-6 months old) and adult cynomologus monkeys (9-15 years old). This technique, originally described by the laboratory of Dr. Lichtman (Gan et al., 2000), is well suited for the study of dendritic branching and dendritic spine morphology and can be combined with traditional...

This protocol describes a simple method for isolating and culturing primary mouse cerebral granule neurons (CGNs) from 6-7 day old pups, efficient transduction of CGNs for loss and gain of function studies, and modelling NMDA-induced neuronal excitotoxicity, low-potassium-induced cell death, DNA-damage, and oxidative stress using the same culture model.
