Oxygen Pressure Modulation

Oxygen pressure modulation is the controlled adjustment of oxygen partial pressure in a gas, liquid, or enclosed environmental system to regulate oxygen availability and its effects. Because oxygen partial pressure influences the concentration gradient driving diffusion and, in water, the amount of oxygen that can dissolve, raising or lowering it changes local redox conditions and the activity of oxygen-dependent reactions. In environmental research, this approach helps test how organisms, microbial communities, sediments, and treatment systems respond to changing oxygen supply, supporting studies of respiration, nutrient cycling, pollutant transformation, and resilience to fluctuating conditions.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

The laser heated diamond anvil cell combined with synchrotron micro-diffraction techniques allows researchers to explore the nature and properties of new phases of matter at extreme pressure and temperature (PT) conditions. Heterogeneous samples can be characterized in situ under high pressure by 2D mapping and combined powder, single-crystal and multigrain diffraction approaches.

Polymersomes are self-assembled polymeric vesicles that are formed in spherical shapes to minimize Gibb's Free Energy. In the case of drug delivery, more elongated structures are beneficial. This protocol establishes methods to create more rod-like polymersomes, with elongated aspect ratios, using salt to induce osmotic pressure and reduce internal vesicle volumes.

Detecting Reactive Oxygen Species

2023

Reactive oxygen species are chemically active, oxygen-derived molecules capable of oxidizing other molecules. Because of their reactive nature, there are many deleterious effects associated with unchecked ROS production, including structural damage to DNA and other biological molecules. However, ROS can also be mediators of physiological signaling. There is accumulating evidence that ROS play significant roles in everything from activation of transcription factors to the mediation of...
