Thermocycler Amplification

Thermocycler amplification is a laboratory method that uses repeated temperature changes to produce many copies of a selected DNA sequence, making scarce genetic material measurable and analyzable. In polymerase chain reaction (PCR), the instrument cycles through denaturation to separate DNA strands, primer annealing to define target boundaries, and extension, during which a thermostable DNA polymerase synthesizes complementary strands; repeated cycles amplify the target exponentially when reagents and temperatures are appropriately controlled. In genetics, the method supports genotyping, mutation detection, cloning, pathogen identification, and analysis of DNA from limited samples. Its speed and sensitivity make it a foundation of molecular research and diagnostic workflows.

Rapid PCR Thermocycling using Microscale Thermal Convection

We describe a novel method to perform DNA replication via the polymerase chain reaction (PCR). Thermal convection is harnessed to continuously shuttle reagents between denaturing, annealing, and extension conditions by maintaining opposing surfaces of the reactor at constant temperature. This inherently simple design promises to make rapid PCR more accessible.
