Antigen Internalization

Antigen internalization is the uptake of extracellular antigen by immune cells, a process that determines how the immune system detects and responds to pathogens and vaccines. Specialized receptors on dendritic cells, macrophages, or B cells bind antigen and trigger receptor-mediated endocytosis or phagocytosis; vesicles then transport the material for proteolytic processing and loading onto major histocompatibility complex molecules. This pathway supports antigen presentation to T cells, shapes adaptive immunity, and influences infection outcomes, vaccine design, immunotherapy, and studies of host-pathogen interactions.

Cellular Affinity of Particle-Stabilized Emulsion to Boost Antigen Internalization

To rationally design efficient adjuvants, we developed poly-lactic-co-glycolic acid nanoparticle-stabilized Pickering emulsion (PNPE). The PNPE possessed unique softness and a hydrophobic interface for potent cellular contact and offered high-content antigen loading, improving the cellular affinity of the delivery system to antigen-presenting cells and inducing efficient internalization of antigens.
