Blood Culture Isolation

Blood culture isolation is a microbiological method for recovering and identifying microorganisms circulating in the bloodstream, making it essential for diagnosing bacteremia, fungemia, and other serious infections. A patient blood sample is inoculated into culture bottles containing nutrient media and incubated under aerobic or anaerobic conditions; microbial growth generates detectable changes, after which organisms are isolated, identified, and tested for antimicrobial susceptibility. In immunology and infection research, this process links bloodstream infection with host immune responses and helps distinguish true infection from contamination. Results guide targeted treatment, support surveillance of antimicrobial resistance, and improve understanding of sepsis and other invasive diseases.

Isolation and Culture of Pulmonary Endothelial Cells from Neonatal Mice

Here, we describe a protocol for isolation and culture of murine pulmonary endothelial cells. This method comprises mechanic and enzymatic lung tissue dissociation as well as a 2-step purification process using anti-PECAM-1 and anti-ICAM-2 antibodies conjugated to magnetic beads, which produces a pure endothelial cell population of mostly microvascular origin.

The goal of this protocol is to isolate endothelial progenitor cells from umbilical cord blood. Some of the applications include using these cells as a biomarker for identifying patients with cardiovascular risk, treating ischemic diseases, and creating tissue-engineered vascular and heart valve constructs.

Generation and Culture of Blood Outgrowth Endothelial Cells from Human Peripheral Blood

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.
