Collagen Overlay

Collagen overlay is a cell-culture technique that places a collagen gel over adherent cells to recreate key features of a three-dimensional extracellular matrix. A neutralized collagen solution polymerizes into a fibrillar network, surrounding the cells with a porous layer that influences cell shape, migration, signaling, and access to soluble molecules. In immunology and infection research, collagen overlays help model tissue-like environments for studying leukocyte movement, epithelial barrier responses, microbial invasion, and host-pathogen interactions. By providing greater structural context than a standard monolayer, the method can improve analysis of cellular behavior and infection processes under experimentally controlled conditions.

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.

This procedure demonstrates in vivo near IR fluorescence imaging of collagen remodeling activities in mice as well as ex vivo staining of collagens in tissue sections using caged collagen mimetic peptides that can be photo-triggered to hybridize with denatured collagen strands.
