Macrophage Cells

Macrophage cells are innate immune cells that detect, engulf, and process pathogens, damaged cells, and cellular debris, making them central to tissue defense and homeostasis. They recognize molecular patterns through surface and intracellular receptors, then use phagocytosis, degradative enzymes, and antimicrobial molecules to destroy targets while releasing cytokines that coordinate inflammation; macrophages can also present antigens to adaptive immune cells. In immunology and infection research, they provide a model for studying host–pathogen interactions, inflammatory signaling, immune evasion, and tissue repair. Their diverse activation states also inform research into chronic infections, autoimmune disease, cancer, and therapeutic immune modulation.

Differentiation and Polarization of Monocyte-Derived Cells into Macrophage-Like Cells

In this video, we demonstrate a procedure to induce the differentiation and polarization of peripheral blood-derived monocytes into M1 or M2 macrophages. Incubation with granulocyte-macrophage colony-stimulating factor, lipopolysaccharide, and interferon‐gamma yields pro-inflammatory M1 macrophages, while incubation with macrophage colony-stimulating factor and interleukin-4 induces anti-inflammatory M2 macrophage production.

Differentiation of Human-Induced Pluripotent Stem Cells into Macrophages

This video describes an in vitro method for generating macrophages from human induced pluripotent stem cells (iPSCs). Macrophage generation from iPSCs occurs in three stages: (i) aggregation of iPSCs in suspension to form three-dimensional embryoid bodies (EBs) that differentiate into mesodermal cells, (ii) formation of macrophage precursor cells from the differentiated EBs, and (iii) differentiation of the macrophage precursor cells into mature, functional macrophages.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

Here, we detail a methodology for the rapid isolation of mouse intestinal dendritic cells (DCs) and macrophages. Phenotypic characterization of intestinal DCs and macrophages is performed using multi-color flow cytometric analysis while magnetic bead enrichment followed by cell sorting is used to yield highly pure populations for functional studies.

Immunohistochemistry to Study the Relationship Between Macrophages and Infiltrated Immune Cells

This video demonstrates a technique to quantify the relative ratio of macrophages among the total infiltrated immune cell populations in temporal artery biopsy. The immunohistochemical staining of a tissue section affected by autoimmune disease selectively stains the immune cells with specific receptors to visually differentiate them from the rest of the cells.
