Nf Kappa B Activity

NF-kappa B activity is the regulated signaling process by which NF-kappa B transcription factors control genes involved in inflammation, immune defense, cell survival, and stress responses. In resting cells, NF-kappa B remains inactive in the cytoplasm through association with inhibitory IκB proteins; stimulation by microbial components, cytokines, or cellular stress activates IκB kinases, leading to IκB degradation and NF-kappa B translocation into the nucleus. There, it binds specific DNA sequences and alters transcription of inflammatory mediators and immune-response genes. Studying this pathway helps explain host responses to infection and supports research into chronic inflammation, immune disorders, and therapeutic targets.

Here, we present a protocol to quickly and easily measure nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) activation in cell lines expressing NF-κB::luciferase reporter constructs, via measurements of luminescence in the cell lysate. Additionally, gene expression is determined via RT-qPCR isolated from cells infected with Salmonella Typhimurium.

Methods for the manipulation and analysis of NF-κB-dependent adult hippocampal neurogenesis are described. A detailed protocol is presented for a dentate gyrus-dependent behavioral test (termed the spatial pattern separation-Barnes maze) for the investigation of cognitive outcome in mice. This technique should also help enable investigations in other experimental settings.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

Measurement of Larval Activity in the Drosophila Activity Monitor

This report describes a method for measuring Drosophila larval activity using the TriKinetics Drosophila Activity Monitor. The device employs infrared beams to detect movements of up to 16 individual animals. Data can be analyzed to represent motion parameters including rates and the positions of the animals within the assay chambers.
