Pbs Colon Flushing

PBS colon flushing is a sampling technique that rinses the large intestine with phosphate-buffered saline (PBS) to recover luminal contents for immunology and infection studies. The buffered, near-isotonic solution mechanically dislodges mucus, microbes, immune cells, and soluble molecules from the colonic lumen while helping maintain sample integrity during collection. Researchers can then analyze the recovered material to assess intestinal inflammation, microbial burden, immune responses, and host–pathogen interactions. Because the procedure provides a standardized way to collect colonic contents, it supports comparisons across experimental conditions and helps connect changes in the gut environment with systemic or tissue-level immune outcomes.

Source: Madeline Lassche, MSNEd, RN and Katie Baraki, MSN, RN, College of Nursing, University of Utah, UT After peripheral intravenous (IV) access is initiated, it is important to assess and maintain the IV catheter according to institutional policies and nursing standards of practice. The regular assessment of the insertion site and the surrounding areas for signs of complications is necessary to prevent IV catheter complications, including infiltration, phlebitis, infection, extravasation, or...

This video demonstrates a method to record the activity of colonic afferent nerves in an ex-vivo rat colon, cannulated at both ends and infused with saline to increase intraluminal pressure. As pressure increases, the nerve signal rises, indicating its stimulation.

Colonization of the murine nasopharynx with Streptococcus pneumoniae and the subsequent extraction of adherent or recruited cells is described. This technique involves flushing the nasopharynx and collection of the fluid through the nares and is adaptable for various readouts, including differential cell quantification and analysis of mRNA expression in situ.

Isolation of Primary Myofibroblasts from Mouse and Human Colon Tissue

The myofibroblast is an influential stromal cell of the gastrointestinal tract that regulates important physiologic processes in both normal and disease states. We describe a technique that allows for the isolation of primary myofibroblasts from both mouse and human colon tissue, which can be utilized for in vitro experimentation.
