Subcutaneous Tissue Dissection

Subcutaneous tissue dissection is a surgical technique used to expose and separate the connective tissue layer beneath the skin while maintaining access to underlying anatomical structures. It works by carefully opening the skin and following tissue planes with controlled blunt or sharp separation, allowing researchers to isolate regions, collect specimens, or visualize local inflammation without unnecessarily disturbing deeper tissues. In immunology and infection research, this approach supports assessment of immune cell recruitment, edema, tissue damage, and pathogen distribution in subcutaneous models. These observations help connect local host responses with disease progression and evaluate interventions that alter inflammation or microbial spread.

A Technique for Subcutaneous Abdominal Adipose Tissue Biopsy via a Non-diathermy Method

We standardized an abdominal adipose tissue biopsy using a non-diathermy method performed under local anesthesia. Three cases of excessive post-operation bleeding out of 115 operations (2.61%) occurred.We conclude that an abdominal adipose tissue surgical biopsy using a non-diathermy method can be safely applied to healthy men.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.
