Cell Labeling Protocol

A cell labeling protocol is a method for marking specific cells so they can be identified, quantified, sorted, or tracked in medical research. Labels may consist of fluorescent dyes, antibody conjugates that bind cell-surface markers, or genetically encoded reporters that produce detectable signals within living cells. Researchers use microscopy, flow cytometry, and cell-sorting approaches to detect these signals and evaluate cell behavior, distribution, viability, or interactions. In medicine, cell labeling supports immune-cell characterization, disease modeling, drug development, and cell-based therapies, helping investigators follow transplanted cells and assess treatment-related changes in complex biological systems.

Contactless dielectrophoresis (cDEP) achieves sorting and enrichment of particles via their intrinsic dielectric properties. Fluidic electrode channels replace metallic electrodes traditional to DEP, suiting cDEP to non-damaging sterile characterization and sorting of biological particles. We demonstrate how to prepare a cDEP microdevice and conduct cell characterization and sorting experiments.

Cell cycle analysis with 5-ethynyl-2'-deoxyuridine (EdU) and phospho-histone H3 (pH3) labeling is a multi-step procedure that may require extensive optimization. Here, we present a detailed protocol that describes all steps for this procedure including image analysis and quantification to distinguish cells in different cell cycle phases.
