Glaucoma Diagnosis

Glaucoma diagnosis is the clinical process of identifying glaucomatous damage to the optic nerve and corresponding vision loss, often before symptoms become noticeable. Eye-care professionals combine intraocular pressure measurement, optic nerve examination, visual field testing, optical coherence tomography, and gonioscopy to assess pressure, nerve structure, retinal changes, and the drainage angle; elevated pressure is an important risk factor but does not alone confirm disease. Accurate diagnosis distinguishes glaucoma from other causes of optic nerve or visual field abnormalities and supports timely treatment, monitoring, and risk assessment. Early detection can help preserve vision and guide long-term medical, laser, or surgical care.

Glaucoma is a chronic disease with progressive degeneration of optic nerve fibers resulting in decreased visual field. Elevated intraocular pressure is considered the most important and the only treatable risk factor. This manuscript describes a simple, surgeon-friendly, non-incisional technique, named Ultrasound Cyclo Plasty, for reducing intraocular pressure in glaucoma patients.

Source: Bosco, A., et al. In Vivo Dynamics of Retinal Microglial Activation During Neurodegeneration: Confocal Ophthalmoscopic Imaging and Cell Morphometry in Mouse Glaucoma. J. Vis. Exp. (2015) This video demonstrates in vivo imaging of retinal microglia in a mouse model of glaucoma. A transgenic mouse with fluorescent protein-expressing retinal microglia is positioned under an ophthalmoscope. Infrared illumination is used to locate the optic nerve head (ONH), and the focal plane is...

Glaucoma is characterized by damage to retinal ganglion cells. Inducing glaucoma in animal models can provide insight into the study of this disease. Here, we outline a procedure that induces loss of RGCs in an in vivo rat model and demonstrates the preparation of whole-mount retinas for analysis.

A Magnetic Microbead Occlusion Model to Induce Ocular Hypertension-Dependent Glaucoma in Mice

Here, we present a protocol to induce ocular hypertension in the murine eye that results in the loss of retinal ganglion cells as observed in glaucoma. Magnetic microbeads are injected into the anterior chamber and attracted to the iridocorneal angle using a magnet to block the outflow of aqueous humour.
