Intravital Two Photon Microscopy

Intravital two-photon microscopy is a live-tissue imaging technique that visualizes cellular and molecular dynamics inside living organisms, making it valuable for studying disease processes in their native physiological environment. It uses pulsed near-infrared laser light to drive two-photon fluorescence excitation only at the focal point, which limits out-of-focus photodamage and enables imaging deeper below tissue surfaces. In medicine, the method can track immune-cell migration, tumor growth, blood-vessel behavior, and therapeutic responses over time, linking microscopic events to disease progression and treatment effects.

We have developed novel laboratory tools and protocols for intravital imaging acquisition of the thymus. Our technique should help in the identification of “niches” within the thymus where T cell development occurs.

The extracellular matrix undergoes substantial remodeling during wound healing, inflammation and tumorigenesis. We present a novel intravital immunofluorescence microscopy approach to visualize the dynamics of fibrillar as well as mesh-like matrix components with high spatial and temporal resolution using epifluorescence or two-photon microscopy.

Highly Resolved Intravital Striped-illumination Microscopy of Germinal Centers

High-resolution intravital imaging with enhanced contrast up to 120 µm depth in lymph nodes of adult mice is achieved by spatially modulating the excitation pattern of a multi-focal two-photon microscope. In 100 µm depth we measured resolutions of 487 nm (lateral) and 551 nm (axial), thus circumventing scattering and diffraction limits.

We have established a cortical orthotopic glioblastoma model in mice for intravital two-photon microscopy that recapitulates the biophysical constraints normally at play during the growth of the tumor. A chronic glass window replacing the skull above the tumor enables the follow-up of the tumor progression over time by two-photon microscopy.
