Phosphatidylserine Expression

Phosphatidylserine expression refers to the presence and distribution of the membrane phospholipid phosphatidylserine, particularly its exposure on the cell surface, where it influences cell signaling and blood coagulation. In healthy cells, phosphatidylserine is concentrated on the inner leaflet of the plasma membrane; during apoptosis or cellular activation, increased intracellular calcium activates scramblases and inhibits flippases, relocating it to the outer leaflet. In medicine, surface phosphatidylserine serves as a marker of apoptotic cells and activated platelets, supporting research on tissue injury, thrombosis, cancer, immune regulation, and diagnostic or therapeutic strategies targeting abnormal membrane signaling.

Annexin V Binding Assay: A Fluorescence-Based Technique to Identify Apoptotic Erythrocytes via Phosphatidylserine Labeling

In this video, we demonstrate the Annexin V binding assay to measure the degree of eryptosis, erythrocyte programmed cell death, using fluorescent Annexin V staining. Apoptotic erythrocytes demonstrate phosphatidylserine translocation from the inner to the outer leaflet of the cell membrane, causing the fluorophore-tagged Annexin V to bind to the translocated phosphatidylserine, thereby emitting a fluorescent signal.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.
