Muscle Evaluation

Muscle evaluation is the systematic assessment of muscle size, tone, strength, function, and performance to identify weakness, injury, pain, or neuromuscular disease. Clinicians integrate patient history with inspection and palpation, range-of-motion assessment, manual resistance testing, reflexes, and functional tasks such as standing, walking, or climbing; differences in force production and movement can indicate problems in muscle tissue, peripheral nerves, neuromuscular junctions, or the central nervous system. In medicine, these findings guide differential diagnosis, establish baseline function, track recovery after injury or surgery, and help determine when laboratory tests, imaging, or electrodiagnostic studies are warranted.

Here we present a detailed protocol to detect both senescent and pluripotent stem cells in the skeletal muscle upon injury while inducing in vivo reprogramming. This method is suitable for evaluating the role of cellular senescence during tissue regeneration and reprogramming in vivo.

We describe an in vivo protocol to measure dorsiflexion of the foot following stimulation of the peroneal nerve and contraction of the anterior crural compartment of the rat hindlimb. Such measurements are an indispensable translational tool for evaluating skeletal muscle pathology and tissue engineering approaches to muscle repair and regeneration.
