Branching Structure Analysis

Branching Structure Analysis is a quantitative approach for describing how biological structures divide into interconnected branches, with particular importance in neuroscience for characterizing neuronal morphology. In practice, researchers reconstruct dendritic or axonal arbors from microscopy images and measure features such as branch points, segment lengths, branching angles, and spatial distribution, often using Sholl analysis to assess complexity across concentric regions. These measurements reveal how neurons organize processes for receiving and transmitting signals. Comparing branching patterns across brain regions, developmental stages, disease models, or experimental treatments can link structural changes to connectivity, cellular function, and neural circuit development.

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure.
