Dopamine Transporter Kinetics

Dopamine transporter kinetics describes how the dopamine transporter (DAT) controls the rate and capacity of dopamine clearance from synapses, a process central to neuronal signaling. DAT uses sodium and chloride ion gradients to drive dopamine reuptake into presynaptic neurons, and kinetic parameters such as the Michaelis-Menten constant (Km) and maximum transport rate (Vmax) quantify transporter affinity and activity. In neuroscience, these measurements help distinguish changes in dopamine signaling caused by transporter regulation, genetic variation, psychoactive drugs, or disease. Kinetic analysis supports research on reward, motivation, motor control, addiction, and disorders involving dopaminergic dysfunction, while informing the development and evaluation of DAT-targeting therapeutics.

Creating Dynamic Images of Short-lived Dopamine Fluctuations with lp-ntPET: Dopamine Movies of Cigarette Smoking

We present a novel PET imaging approach for capturing dopamine fluctuations induced by cigarette smoking. Subjects smoke in the PET scanner. Dynamic PET images are modeled voxel-by-voxel in time by lp-ntPET, which includes a time-varying dopamine term. The results are 'movies' of dopamine fluctuations in the striatum during smoking.

Single Cell Measurement of Dopamine Release with Simultaneous Voltage-clamp and Amperometry

The amperometric technique measures dopamine release from a single cell by detecting the oxidative current produced by spontaneous dopamine oxidization. Simultaneous voltage clamp and amperometry methodology reveal the mechanistic relationship between the overall "activity" of dopamine transporter and the regulatory role of this activity on the reverse transport of dopamine.

Fast-scan cyclic voltammetry can monitor in vivo dopamine neurotransmission in the context of drugs, disease, and other experimental manipulations. This work describes the implementation of QNsim1.0, a software to model electrically stimulated dopamine responses according to the quantitative neurobiological model to quantify estimates of dopamine release and reuptake dynamics.
