Glioma Metastasis

Glioma metastasis is the dissemination of malignant glial tumor cells from a primary brain tumor to new locations within the central nervous system or, more rarely, beyond it, making it an important challenge in neuroscience and oncology. Glioma cells invade surrounding brain tissue, migrate along blood vessels and white-matter tracts, and may enter cerebrospinal fluid or the bloodstream; successful dissemination requires survival, movement, and colonization of a new microenvironment. Although extracranial spread is uncommon, metastatic glioma can involve the spinal cord or distant organs. Studying these processes supports research into tumor progression, diagnostic biomarkers, therapeutic resistance, and treatments that limit invasion and recurrence.

This video demonstrates the assessment of neuronal electrical activity in a co-culture of hiPSC-derived cortical glutamatergic neurons and glioma cells using a microfluidic device with multielectrode arrays.

Co-culturing Glutamatergic Neurons and Pediatric High-Grade Glioma Cells

This video showcases a technique for co-culturing glutamatergic neurons and pediatric high-grade glioma (pHGG) cells within a microfluidic device. Human induced pluripotent stem cells are introduced into the microfluidic system and incubated to promote their differentiation into mature cortical glutamatergic neurons. Following this, the pHGG cells are introduced onto the mature neurons, establishing a co-culture and enabling interactions between neurons and pHGG cells.

Advanced Animal Model of Colorectal Metastasis in Liver: Imaging Techniques and Properties of Metastatic Clones

The ability of metastatic clones to colonize distant sites depends on their proliferation capacity and/or their ability to survive in the host microenvironment without significant proliferation. Here, we present an animal model that allows quantitative visualization of both types of liver colonization by metastatic clones.

Isolation of Nuclei from Fresh Frozen Glioma Tissues: A Method to Obtain Intact Nuclei from Glioma Tumor Samples

This video describes the protocol to isolate single-nuclei from fresh frozen glioma tissue. The isolated single-nuclei can be used as specimens for sequencing techniques such as single-nuclei RNA (snRNA) sequencing and single-nuclei Assay for Transposase-Accessible Chromatin (snATAC) sequencing to obtain deeper insights into inter- and intra-tumor heterogeneity observed in glioma of the central nervous tissues.
