Imaging Agent

An imaging agent is a chemical substance or biological probe that enhances, creates, or targets a detectable signal, enabling researchers to visualize structures, processes, or molecular activity in living systems. In neuroscience, these agents may cross or interact with the blood-brain barrier and produce signals through mechanisms such as radioactive decay, magnetic susceptibility changes, or selective binding to receptors and transporters. Used with modalities including positron emission tomography, magnetic resonance imaging, and optical imaging, imaging agents help map brain anatomy, neurotransmitter systems, metabolism, inflammation, and disease-related changes. They support diagnosis, drug development, and investigations of brain function and pathology.

Preparation and In Vitro Characterization of Dendrimer-based Contrast Agents for Magnetic Resonance Imaging

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.

We demonstrate how to perform macrophage MR imaging using ultrasmall superparamagnetic contrast agent (USPIO) in septic arthritis, allowing an initial and longitudinal in vivo non-invasive evaluation of macrophages infiltration and an assessment of therapy efficacy.
